Moura C., Tiba M.R., Silva M.J. & Leite D.S. 2013. Identification of new flagellin-encoding fliC genes in Escherichia coli isolated from domestic animals using RFLP-PCR and sequencing methods. E-mail: [email protected] Identification of Escherichia coli requires knowledge regarding the prevalent serotypes and virulence factors profiles allows the classification in pathogenic/non-pathogenic. However, some of these bacteria do not express flagellar antigen in vitro. In this case the PCR-restriction fragment length polymorphism (RFLP-PCR) and sequencing of the fliC may be suitable for the identification of antigens by replacing the traditional serology. We studied 17 samples of E. coli isolated from animals and presenting antigen H nontypeable (HNT). The H antigens were characterized by PCR-RFLP and sequencing of fliC gene. Three new flagellin genes were identified, for which specific antisera were obtained. The PCR-RFLP was shown to be faster than the serotyping H antigen in E. coli, provided information on some characteristics of these antigens and indicated the presence of new genes fliC.

Identification of putative new Escherichia coli flagellar antigens from human origin using serology, PCR-RFlP and DNA sequencing methods.

Escherichia coli has been isolated frequently, showing flagellar antigens that are not recognized by any of the 53 antisera, provided by the most important reference center of E. coli, The International Escherichia and Klebsiella Center (WHO) of the Statens Serum Institute, Copenhagen, Denmark. The objective of this study was to characterize flagellar antigens of E. coli that express non-typeab...

Sequencing and comparative analysis of flagellin genes fliA and fliB in bovine Clostridium chauvoei isolates

Background: Clostridium chauvoei is the etiological agent of blackleg as an endogenous infection in cattle.  Flagella have been known to play a critical role in the protective immunity of animals to clostridial infections. C.chauvoei has two copies of fliC gene, namely fliA and fliB. OBJECTIVES: The aim of this study was the determination and nucleotide sequence analysis of both copies of fliC ...

Identification of Biofilm Encoding Genes (agg) in the Escherichia coli Isolates by Multiplex-PCR Method

Introduction: Biofilm is a collection of microbial cells that are irreversibly suppressed and mildly washed away. The aggR gene is located on the main plasmid (pAA), which codes for many actuator factors. The pathogen strains of EAEC have an aggR gene, the gene has been identified in six classes of pathogenic E. coli.   Materials & Methods: In the present study, 60 stool samples from children ...

Can the fliC PCR-restriction fragment length polymorphism technique replace classic serotyping methods for characterizing the H antigen of enterotoxigenic Escherichia coli strains?

In this study, we performed fliC PCR-restriction fragment length polymorphism (RFLP) to investigate whether this technique would be better than classic serotyping for the characterization of the H antigen in enterotoxigenic Escherichia coli (ETEC) strains. We showed that the fliC genes from ETEC strains can be characterized by restriction analysis of their polymorphism. Only one allele of the f...

Abstract Background and Objectives: Efflux pumps are one of the main mechanisms for antibiotic resistance in Escherichia coli strains. The aim of this study was to investigate the relationship between 5 different efflux pump genes; acrA, acrB, emrA, emrB and mdtk and fluoroquinolone resistance in E.coli Isolated from patients with urinary tract infections in Qom. Methods: In this descriptive cr...
